The E/LE series Focused-ultrasonicators are based on Covaris Adaptive Focused Acoustics (AFA) technology. These instruments are designed to provide researchers an integrated standalone tool to be used in a laboratory environment. The instrument is ideal for those who work with tissue homogenization, cell lysis, DNA or chromatin shearing, and other sample preparation steps. The system is comprised of the Treatment System which delivers the acoustic energy to the sample, the Safety System which protects users from inadvertently contacting the high intensity acoustic energy, and the Computer and Application Software which provides users an interface to control the device. The Covaris Process uses adaptive focused acoustic energy to precisely control cavitation and acoustic streaming within the sample treatment vessel in a non-contact, isothermal fashion. Focused acoustic energy is generated and delivered to individual samples. The mechanical energy imparted on the sample results in a controlled series of compression and rarefaction events.

Technical Notes

LE220: The unit was connected through a USB cable to a laptop with Sonolab 7.3 installed . The water was cooled to 10 °C with the circulator and conditioned with WCS. The unit was homed (initialized) successfully. We then performed degassing until the degassing timer ran down after 60 min. Degassing indicator turns green. A sample of water and carbon was added in a microtube and loaded onto the rack. Door lock mechanism works. The sample was treated successfully, as the water and carbon mixed right away once the treatment starts. See video and photos for details. The instrument was shut down according to the instruction in the menu. Water in the degassing pump was drained.

Polyscience circulator: tested all buttons and passed. We used water as the cooling fluid since there is no need to cool to freezing (use polycool if below freezing temperature is desired). Water was cooled to 5 °C.

WCS water conditioning system: powers on. Hour meter starts running. Only 26 hours of usage.

Laptop: latitude 3470 with Windows 10. Sonolab 7.3 installed. Well plate definition installed. 8gb of ram and 240 gb of SSD.

Water in all of chiller hose was drained.

The system was commissioned in 2014, judging from the document included, and last serviced in 2016.
